Description

Description: This assay employs a two-site sandwich ELISA to quantitate XRCC4 in samples. An antibody specific for XRCC4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any XRCC4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XRCC4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XRCC4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: X-Ray Repair Cross-Complementing 4 functions together with DNA ligase IV and the DNA-dependent protein kinase in the repair of DNA double-strand break by non-homologous end joining and the completion of V(D)J recombination events. The non-homologous end-joining pathway is required both for normal development and for suppression of tumors.
This gene functionally complements XR-1 Chinese hamster ovary cell mutant, which is impaired in DNA double-strand breaks produced by ionizing radiation and restriction enzymes. This gene contains 8 exons, and alternative transcription initiation and alternative splicing generates several transcript variants
